S11 surfactant/Surfynol 485
Surfynol 485 is a non-ionic surfactant based on the ethoxyl compound of acetylenediol, which is prepared by the addition of 85% ethylene oxide (30 mol EO) and Surfynol 104 (tetramethyldecynediol). Its unique molecular structure gives it ultra-low dynamic/static surface tension, high hydrophilicity (HLB=17) and non-hemolytic properties, which is a key functional aid for in vitro diagnostic reagents to achieve efficient wetting and stabilization of latex systems and optimize whole blood detection performance.
Action mechanism
Ultra-low surface tension: rapidly reduce the liquid-solid interfacial tension, and promote the uniform spread and penetration of reagents on hydrophobic surfaces (such as cellulose nitrate film and plastic microporous plates).
Non-hemolytic: Acts gently on erythrocyte membrane to avoid hemolysis interfering with optical detection (such as whole blood direct chromatography or colorimetry).
Enzyme compatibility: does not destroy enzyme activity, ADAPTS enzyme reaction system (such as ELISA, chemiluminescence), improve the stability of detection.
Low foam and defoaming: inhibit foam formation in the process of reagent mixing or automatic sample addition to ensure the accuracy of signal reading.
Latex particle stabilization: Through the synergistic effect of steric hindrance and electrostatic repulsion, latex/colloidal gold accumulation is prevented and turbidity signal uniformity is improved.
Application scenario
Immunochromatographic strip: Added to the sample pad treatment solution to accelerate whole blood/serum penetration, inhibit red blood cell retention, and reduce background interference.
Latex enhanced immunoturbidimetry: Premixed in latex microsphere suspension to enhance particle dispersion, improve detection sensitivity and linear range.
Whole blood direct detection reagent: as a component of red blood cell adsorption material, gentle separation of plasma and cells, eliminating high HCT sample deviation.
Microfluidic chip wetting agent: applied to hydrophobic microchannels to optimize blood flow uniformity and avoid cell blockage or bubble retention.
Enzyme reaction system stabilizer: enzyme preservation solution is added to reduce enzyme inactivation during lyophilization/resolution process and extend the reagent validity.
General use range: 0.005%-0.5% (0.01%-0.1% commonly used in diagnostic reagents).
Initial test: It is recommended to start from **0.05%** gradient optimization (such as 0.01%, 0.05%, 0.1%, 0.2%), and evaluate the following indicators:
Wetting rate: the diffusion time of the liquid on a solid phase carrier (such as the chromatographic speed of the test strip).
Signal uniformity: dispersion of latex particles or color region consistency.
Background cleanliness: Red blood cell residue or nonspecific adsorption.
Matters needing attention
Solubility: the solubility in water is >1%, but it needs to be slowly stirred to dissolve (to avoid the introduction of bubbles by violent vibration).
Temperature sensitivity: Long-term exposure to >40 ° C may lead to molecular chain degradation.
Environmental compliance: No alkyl phenol polyoxyethylene ether (APE) and organic solvents, in compliance with FDA 21 CFR and EU IVDR regulations.
Q&A
Uneven chromatographic velocity: may be insufficient wettability or concentration is too low; It is recommended to increase to 0.1%-0.2%, or optimize the sample pad pretreatment process.
Fluctuation of latex signal: may be insufficient particle dispersion; It is recommended to increase Surfynol 485 to 0.05%-0.1% to extend the ultrasonic dispersion time.
Enzyme activity decreased: it may be the competitive combination of surfactant and enzyme; It is recommended to reduce the concentration to less than 0.01% or replace it with a more inert agent (such as Poloxam 188).
Reagent residue crystallization: low temperature storage may lead to precipitation; It is recommended to return to room temperature and swirl well, and filter if necessary.
