Surfynol 465 is a non-ionic dienestar surfactant based on ethoxyl dienediol compounds, belonging to the same family as Surfynol 485, but with a lower dynamic surface tension and an optimized hydrophilic to hydrophobic balance (HLB≈15). Its low vesicles, non-hemolytic and mild effect on red blood cells make it the core functional auxiliary for rapid wetting and stabilizing latex system and direct detection of whole blood in vitro diagnostic reagents.

 

Action mechanism

Ultra-low dynamic surface tension: rapidly reduce the liquid-solid interfacial tension and accelerate the penetration and spread of reagents on dense or hydrophobic surfaces (such as cellulose nitrate membranes and plastic microporous plates).

Red blood cell compatibility: inhibit red blood cell rupture or adhesion, adapt to whole blood direct detection (such as chromatographic strips, microfluidic chips), eliminate high HCT sample deviation.

Latex particle stabilization: Through hydrophobic chain adsorption and hydrophilic chain steric hindrance, latex/colloidal gold accumulation is prevented, and turbidity signal uniformity is improved (e.g. CRP detection).

Low foam and anti-interference: Reduce foam formation in reagent mixing or automatic sample addition, avoid optical signal fluctuations (such as colorimetry, fluorescence detection).

Wide chemical compatibility: Tolerance pH 3-10, high salt and organic solvents (such as ethanol, isopropyl alcohol), suitable for complex reaction systems.

 

Application scenario

Whole blood chromatographic test strip: added to the sample pad treatment solution, accelerate whole blood penetration and block red blood cell retention, improve detection sensitivity and specificity.

Latex enhanced immunoturbidity: Premix in latex microsphere suspension to enhance particle dispersion and optimize turbidity signal of antigen-antibody complex.

Microfluidic chip wetting agent: applied to hydrophobic microchannels to optimize blood flow uniformity and avoid cell blockage or bubble retention.

Red blood cell adsorption material: As a red blood cell filtration membrane coating, gentle separation of plasma and cells, reduce hemolysis interference (such as POCT detection).

Enzyme reaction system synergist: enzyme preservation solution is added to reduce enzyme inactivation during lyophilization/resolution process and extend the reagent validity.

 

General use range: 0.005%-0.3% (0.01%-0.1% commonly used in diagnostic reagents).

 

Matters needing attention

Solubility: the solubility in water is >1%, but it needs to be slowly stirred to dissolve (to avoid the introduction of bubbles by violent vibration).

Temperature sensitivity: Long-term exposure to >50 ° C may lead to molecular chain degradation.

Environmental compliance: no APE (alkyl phenol polyoxyethylene ether) and organic solvents, in accordance with FDA 21 CFR and EU IVDR regulations.

 

Q&A

Too fast chromatographic speed leads to weak signal: it may be too strong wettability or too high concentration; It is recommended to reduce to 0.01%-0.05%, or optimize the membrane pretreatment process.

Uneven dispersion of latex particles: may be insufficient ultrasonic dispersion or concentration; It is recommended to extend the ultrasound time (10-15 minutes), or increase it to 0.03%-0.05%.

Reagent residue crystallization: low temperature storage may lead to precipitation; It is recommended to return to room temperature and swirl well, and filter if necessary.

Partial inhibition of enzyme activity: it may be that surfactants compete with enzymes for binding sites; It is recommended to reduce the concentration to less than 0.01% or replace it with an enzyme compatibility agent (such as Poloxam 188).

The dynamic surface tension of Surfynol 465 is lower than that of Surfynol 485.

Surfynol 465 HLB≈15 (more balanced wettability); And Surfynol 485 HLB=17 (higher hydrophilicity)

Surfynol 465 is more suitable for superhydrophobic surface treatment and high viscosity sample penetration. Surfynol 485 is more suitable for rapid water phase wetting and protection of sensitive enzyme system
