The protein desalination centrifugal column is an improved glucan desalination device. Its working principle mainly utilizes the molecular sieve effect of the glucan gel with a network structure to separate substances based on their different molecular sizes. The retention rate of salt and small molecules by the protein desalination column is over 95%, with almost equal volume recovery. The sample will not be diluted, and it can perform protein desalination excellently while maintaining an extremely high recovery rate. This product does not require column mounting and can replace the time-consuming traditional dialysis treatment to achieve the purpose of rapidly purifying proteins/replacing protein buffers. Just a few simple centrifuges are needed to provide trouble-free desalination and buffer exchange for samples ranging from 100μL to 3mL.

Operation steps
Balance the desalination column
a. Remove the plug at the bottom of the centrifugal desalination column. First, slightly twist it in one direction, then twist it in the other direction, and then put it back into the collection tube. Centrifuge at 1000×g for 1 minute and pour out the protective solution in the collection tube.
b. Place the centrifuge column back into the collection tube, add 250μl of equilibrium solution (the buffer solution to be replaced after sample desalination) to the centrifuge column, let it stand until the equilibrium solution has fully permeated the packing material, then centrifuge at 1000×g for 1 minute. Repeat this process three times to remove the collection solution.
2. Sample processing
Place the centrifugal column into a clean EP tube, add 100μl to 200μl of the sample, let it stand for 3 minutes, then centrifuge at 1000×g for 1 minute. The solution in the EP tube is the desalinated sample.
3. Regeneration and preservation of the desalination column
Add 300μl of 0.5M NaOH solution to the used desalination column, let it stand for 2 minutes, centrifuge at 1000×g for 1 minute, and repeat twice. Then add 300μl of deionized water, let it stand for 2 minutes, centrifuge at 1000×g for 1 minute, and repeat this process three times. After processing the desalination column, add 20% ethanol solution, place a red plug at the lower end, tighten the cap, and store it at 2 to 30 degrees Celsius.
Precautions
1.The sample processing volume should be within the column processing range. Excessive processing will lead to incomplete desalination, while insufficient processing will result in a decrease in sample recovery rate.
2. The packing material may lose water and shrink in high-concentration alcohol solutions or saturated salt solutions. Do not pass the above solutions through the column.
3. When adding samples, try to add them evenly to the center of the tube.
4. When the sample concentration is too low, it is necessary to first concentrate the sample to the required volume or concentration.
