Phosphatase Inhibitor Cocktail 3

Application.
The mixture of phosphatase inhibitors can increase the yield of intact proteins by adding inhibitors to enzymes that can modify proteins present in cell extracts. The product has been tested on animal tissues including human placenta and bovine liver, as well as extracts from A431, CHO and U937 cells. It has optimized and tested L-isozymes of alkaline phosphatase and serine-threonine protein phosphatase for measurement by pNPP.

Biochemical / physiological effects.
The mixture contains independent components with specific inhibitory properties. Cantharidin can inhibit protein phosphatase 2A. Levamisole p-oxalate can inhibit the L-isozyme of alkaline phosphatase. Calyx sponge carcinogen A can inhibit protein phosphatase 1 and 2A.

Warning.
The product can be stable for four years at the recommended storage temperature of 2-8 °C after supply.

Points for attention in preparation.
The mixture is provided in a transparent solution soluble in dimethyl sulfoxide. It is recommended that every 500 mg of protein extracted from tissues or cells be diluted with a mixture of 1 mL. A final concentration of 1% (v / v) is commonly used, which can inhibit phosphatase activity in 27000 × g supernatants of human placenta and bovine liver and 100000 × g supernatants of A431, CHO and U937 extracts.
