Application.
The mixture of phosphatase inhibitors can increase the yield of intact proteins by adding inhibitors to enzymes that can modify proteins present in cell extracts. The product has been optimized and tested for tyrosine protein phosphatase, acid and alkaline phosphatase. It has tested cell extracts from animal tissues including bovine liver and human placenta and rabbit muscle cell extracts.
Biochemical / physiological effects.
The mixture contains independent components with specific inhibitory properties. Sodium orthovanadate can inhibit a variety of ATP enzymes, protein tyrosine phosphatase and other phosphotransferases. Sodium molybdate can inhibit acid and phosphoprotein phosphatase. Sodium tartrate can inhibit acid phosphatase. Imidazole can inhibit alkaline phosphatase.
Warning.
The product can be stable for two years at the recommended storage temperature of 2-8 °C after supply.
Points for attention in preparation.
The mixture is provided in a transparent solution. It has been aseptically filtered through the 0.2 μ m membrane in the aseptic filled bottle. One milliliter would inhibit phosphatase activity at a concentration of about 5 mg/mL found in 100000 x g supernatants from human placenta, bovine liver, rabbit muscle, A431 or Jurkat cell extracts.
