When preparing the storage solution, 10 mg/mL should be dissolved in water and filtered and sterilized.
Biochemical / physiological behavior.
Mechanism of action: spectinomycin inhibits protein synthesis and extension by binding to bacterial 30s ribosomal subunit and interfering with peptide tRNA translocation. Resistance mechanism: rpsE (ribosomal protein S5 gene) mutation can prevent spectinomycin binding. Aminoglycoside-3-adenosine transferase gene (aadA) can also make bacteria resistant. Antibacterial spectrum: spectinomycin can fight against Gram-negative and Gram-positive bacteria.
Warning.
The solution can be stored at 2-8 °C for several weeks or longer at-20 °C.
Application.
Spectinomycin is a kind of aminocyclohexol antibiotics, which comes from Streptomyces magnificent. It has been shown to have a wide range of uses, including the treatment of acute gonococcal urethritis and cervicitis, labeling cell layers to monitor cell fate during leaf development, as a selective marker in plant-related transformation systems containing the marker gene Spcr, studying bovine respiratory infections caused by Pasteurella multocida and Mansteurella haemolyticus, and plants that produce plastids that encode RNA polymerase deficiency on MS Agar medium. Spectinomycin can also be used to amplify plasmids with low copy number. It is recommended to be used in cell culture at 7.5-20 mg/L.
