Biochemical / physiological behavior.
Mode of action: polymyxin B sulfate partially binds to the lipid An of bacterial lipopolysaccharide and destroys the plasma membrane by inducing enough pores to make nucleotides leak from the bacterial wall and make nucleotides leak into the bacterial cell wall. This can break the semi-permeability of the cell plasma membrane. Antibacterial spectrum: it has bactericidal effect on most Gram-negative bacilli, including Escherichia coli, fungi and Gram-positive bacteria.

Warning.
The product should be stored away from light at 2-8 °C. After stored under these conditions for 3 years, no change of the sample was observed. The reserve liquid should be aseptically filtered and stored at 2-8 °C and stable for 5 days at 37 °C.
General description.
Chemical structure: polypeptide polymyxin B is a member of polymyxin antibiotics, obtained from different strains of polymyxomycetes and related species. It contains a cyclic heptapeptide unit and a tripeptide side chain, which is N-acylated by fatty acyl residues. Polymyxin B sulfate is commonly used in the treatment of gram-negative bacterial infections.
Application.
Polymyxin B sulfate is used as a LPS (lipopolysaccharide) inhibitor to treat macrophages of BAC-1.2F5 mice as a positive and negative control of microbial germicidal concentration (MCC) determination.
Points for attention in preparation.
Polymyxin B sulfate dissolves in water at 50 mg/mL to form a slightly turbid, colorless to yellow solution. Its solubility in any organic solvent is very small. For example, its solubility in ethanol is 0.115mg/ml.
