Polymyxin B sulfate salt

Warning.
The product should be stored in 2-8 °C dark conditions. After three years of storage under these conditions, no change was observed in the preserved samples. The reserve solution should be aseptically filtered and stored at 2-8 °C. It can be stable for 5 days at 37 °C.

Points for attention in preparation.
Polymyxin B sulfate can be dissolved in water at 50 mg / mL, resulting in a very slightly turbid, colorless to yellow solution. It has little solubility in any organic solvent. For example, its solubility in ethanol is 0.115mg / mL.

Biochemical / physiological behavior.
Mode of action: polymyxin B sulfate binds to the lipid A part of bacterial lipopolysaccharide, which destroys the plasma membrane by inducing enough pores to allow nucleotides in the bacterial wall to leak. This destroys the permeability of the plasma membrane. Antibacterial spectrum: it has bactericidal effect on most Gram-negative bacilli, including Escherichia coli, as well as most fungi and Gram-positive bacteria.

Application.
Polymyxin B sulfate is a strong cationic cyclic polypeptide antibiotic derived from the fermentation of polymyxobacterium (Bacilus polymyxa). It is a mixture of B1 and B2 sulfates. This product has been used in clinical treatment of urinary tract, meningeal and blood infections caused by susceptible strains of Pseudomonas aeruginosa (Pseudomonas aeruginosa). It is also used to study multidrug resistant pathogens as an immobilization reagent for endotoxin removal and to induce pore formation in the cortical cell membrane of excised sorghum roots.
