General description.
PIPES is one of the ethanesulfonate buffer salts, which was first developed by Good et al to meet certain criteria: median pKa, maximum water solubility and minimum solubility in all other solvents, minimum salt effect, minimum temperature variation of pKa, chemical and enzymatic stability, minimum absorption in the visible or ultraviolet spectrum, easy to synthesize. Because its pKa at 37 ℃ is close to physiological pH,PIPES, it can be used in cell culture.
Points for attention in preparation.
The buffer can be prepared by adding alkaline solution to the PIPES free acid and titrating to the appropriate pH value, or by using equal molar sodium salt solution and disodium salt solution to titrate to the appropriate pH value.
