The denatured polyacrylamide gel electrophoresis was tested.
Application.
Used to dissolve and denature proteins for denaturation-PAGE. Most proteins bind to SDS at a ratio of 1.4 to 1. Due to the negative charge of dodecyl sulfonate, the SDS- complexes of various proteins are negatively charged with the same density. Compared with the total negative charge provided by the bound SDS, the original charge of the protein becomes negligible. Because the charge-mass ratio of each protein is basically the same, the mobility of protein-SDS complexes in gel electrophoresis is only based on their size.
