Application.
TCEP is an effective reductant for protein analysis. As a non-mercaptan reductant, it avoids the inherent toxicity of mercaptan compounds. It can destroy the botulinum neurotoxin B heavy chain / light chain complex bound by a single disulfide bond, which is responsible for the endocytosis of the toxin and ultimately leads to toxicity. Because the disulfide bond coupling subunit is characteristic of many toxins (such as ricin, snake venom and all BoNT serotypes), it can be used as a rescue prophylaxis in the case of toxin administration. Tris (2-carboxyethyl) phosphine (TCEP) can be used in several downstream applications, including SDS-PAGE, mass spectrometry, labeling and modification of cysteine-containing compounds with cysteine-specific tags. It prevents the oxidation of protein samples, which makes it a useful buffer because it helps maintain the activity of the enzyme. It has been used for the reduction and determination of glutathione.
General description.
Tris (2-carboxyethyl) phosphine (TCEP) is very effective in cracking disulfide bonds in aqueous solutions. In proteins, TCEP reduces disulfide bonds and keeps other functional groups intact. Unlike other trialkylphosphines (tributylphosphine), it is soluble in water and has no odor. It is also less toxic than 2-mercaptoethanol. These advantages make it better than other reducing agents.
