Points for attention in preparation.
The product is soluble in water, methanol and ethanol, but insoluble in ether and hydrocarbons. When heated, the product produces a transparent orange solution, which is dissolved in 4 mL 95% ethanol at 50 mg to obtain a transparent yellowish green solution. Tetracycline can be quickly destroyed by alkali metal hydroxide solution, and the static solution becomes turbid due to hydrolysis and precipitation. The potency of tetracycline in solutions with pH less than 2 will be reduced.

Application.
Tetracycline is a broad-spectrum polyketone antibiotic that is used clinically to treat bacterial infections such as Rocky Mountain typhus, tick fever, Q fever and Bochen's disease, as well as upper respiratory tract infections and acne. It has been used to study multidrug resistance and potential side effects, including acute pancreatitis. It is suggested that the concentration of 10 mg / L should be used in cell culture.

Warning.

Biochemical / physiological behavior.
Mode of action: tetracycline passively diffuses through proton channels in the cell membrane, binds to 30s ribosomes, and inhibits protein synthesis by preventing aminoacyl tRNA from entering the receptor site of mRNA- ribosomal complex. It also binds to the 50s ribosomal subunit of bacteria, changes the morphology of the cell membrane and leads to the leakage of intracellular contents. This inhibitory effect can be reversed by washing, indicating that it is a reversible binding antibiotic, not an irreversible binding drug. Drug resistance model: this effect is inactivated due to the loss of cell wall permeability. Antibacterial spectrum: includes a wide range of antibacterial activities against Gram-positive and Gram-negative bacteria.
