Application.
PMSF is usually used in cleavage buffers to help preserve needed proteins by inhibiting proteases that degrade proteins after lysis in cells or tissues during protein separation and preparation. PMSF is unstable in aqueous solution. Therefore, the storage solution of PMSF is usually prepared with anhydrous organic solvents (such as 100% ethanol or anhydrous isopropanol). The main features and advantages of PMSF include inhibition of serine proteases, such as trypsin and chymotrypsin, as well as inhibition of cysteine proteases (reduction of mercaptan is reversible) and mammalian acetylcholinesterase toxicity is lower than DFP. But it is not as good as the effective concentration of DFP when the pH value of mM is 7.5 and the half-life is 1 hour. Phenylmethanesulfonyl fluoride has been used as a component of radioimmunoprecipitation buffer in western blotting analysis. It is also used as a component in the lysis buffer of RAW264.7 cells in Western blotting analysis. Phenylmethanesulfonyl fluoride has been used in the following applications: cell fractionation. As a supplement in nuclear protein extraction. Cholesterol esterase (CE) and pseudocholinesterase (PCE) inhibitors. It is used for blood collection before centrifugation to quantify plasma ANP levels.

General description.
Benzenesulfonyl fluoride (PMSF) is a widely used serine protease inhibitor, which has a good inhibitory effect on chymotrypsin, thrombin and trypsin. PMSF acts as an inhibitor by sulfonating the hydroxyl residues of serine residues at the serine protease reaction site.

Biochemical / physiological behavior.
The intake of PMSF will produce analgesic effect independent of its anticholinesterase effect and prolong the analgesic effect of central administration of β-endorphin.
