Application.
It can be used in combination with E64Murd and Leupeptin A to inhibit the degradation of autophagy substrate in autophagy. For this application, the working concentration is usually between 1 μ M and 10 μ M.
Biochemical / physiological behavior.
Gastrostatin An is an inhibitor of acid protease (aspartyl peptidase). It can form a 1:1 complex with proteases such as pepsin, renin, cathepsin D, bovine Chymosin and protease B (Aspergillus Niger). The inhibitor is highly selective and does not inhibit mercaptan protease, neutral protease or serine protease. Soluble γ-secretase and retroviral protease could also be inhibited by gastrostatin A. It has been used to identify proteases from many sources.
Points for attention in preparation.
Pepsin inhibitors can be dissolved at 1 mg/ml in a methanol solution containing 10% (v / v) acetic acid (methanol: acetic acid ratio 9:1). Acetic acid may be necessary when dissolving the peptide with methanol or DMSO. It can be dissolved in ethanol at 1-2 mg/ml, but it may require heating to dissolve completely. Pepsin inhibitor solution can be heated to up to 60 ℃ without causing peptide decomposition. 1 mg/ml reserve solution can be stored stably at 4 ℃ for at least one week. The 1 mM solution with methanol or DMSO as solvent can be stably stored at-20 ℃ for several months. If the solution turns dark yellow, it means the reagent is hydrolyzed. The solution with a working concentration of 1 μ M can be kept stably at room temperature for at least one day.
