Principle.
β-galactosidase can decompose lactose into galactose and glucose. β-galactosidase is not lactose-specific and can hydrolyze simple galactosides. The hydrolysis of 2-nitrophenyl β-D-glucopyranoside led to the release of galactose and yellow chromogenic compounds. The test substrate does not rely on inducing or constituent permeable enzymes to enter the cell, so the reaction is rapid and occurs within 24 hours.

ONPG (2-nitrophenyl β-D-galactoside) is the colorimetric substrate of β-galactosidase.

Molecular biological water can be used to prepare a reserve solution with a concentration of 3mg / ml. Alternatively, a reserve solution of about 20.5mg / ml can be prepared using 100mM sodium phosphate buffer (pH7.3). It may take a little heating to completely dissolve the product.
