Application.
Thiazolyl tetrazolium bromide (MTT) can be used to measure cell proliferation. MTT produces a yellowish solution, which is transformed into dark blue, water-insoluble MTT- by mitochondrial dehydrogenase in living cells. The blue crystal was dissolved with acidified isopropanol and the strength was measured by colorimetry under 570nm. MTT has been used as a histochemical / cytochemical reagent and for the detection of NAD. Because the cyanide trap used binds to cations, MTT cannot be used to detect the ADP ligase system in tissue. MTT is rapidly reduced to methylene and can chelate with nickel, copper and cobalt; cobalt chelates have been used in oxidation systems.
