Points for attention in preparation.
Kanamycin sulfate was dissolved in water at 50 mg/Ml to obtain a clear solution. It is almost insoluble in ethanol, acetone, chloroform, ether and ethyl acetate. The Ph of 1% aqueous solution is 6.5 to 8.5. The aseptic solution can be prepared by aseptic filtration through 0.2 μ m filter.
Application.
Kanamycin sulfate from Streptomyces kanayi (Streptomyces kanamyceticus) has been used in tissue culture.
Warning.
The solution can be stable for about 5 days at 37 °C. The reserve solution can be stored for a long time at 2-8 °C.
Biochemical / physiological behavior.
Mode of action: the product inhibits ribosomal translocation and causes error coding by binding to 70s ribosomal subunits. Resistance model: aminoglycoside modifying enzymes (including acetyltransferase, phosphotransferase, nucleotide transferase) can change this antibiotic and prevent its interaction with ribosomes. Antimicrobial spectrum: kanamycin sulfate is effective against gram-negative and gram-positive bacteria and mycoplasma.
General description.
Kanamycin sulfate is a broad-spectrum aminoglycoside antibiotic from Streptomyces (Streptomyces kanamyceticus). It is used as an additive in the culture medium for the isolation of group D streptococci on kanamycin aescin azide Agar and for selecting transformed plant cells containing neomycin phosphotransferase on kanamycin medium. Kanamycin B resistance gene was used as a selector for transformed cells. Used in biotechnology to inhibit protein synthesis. It is suggested that the amount of 100mg/L should be used in cell culture.
