General description.
Chemical structure: aminoglycosides are the most widely used antibiotics targeting ribosomes. Hygromycin B is an aminocyclophenol antibiotic. Streptomyces hygroscopicus has phosphotransferase (HPH) activity, which can phosphorylate hygromycin B, which helps to activate autoimmunity against the toxicity of the drug.
Application.
Hygromycin B antibiotics can be used for the moisture resistance of the following cells: 160u g/ml for 293T cells transfected with plasmid mixture. 150 g/ml for MCF-7ALOX12 cells. MSCV luciferase PGK-hygro plasmid. STAT3 reporter gene HeLa stable cell line (stably transfected with luciferase gene controlled by STAT3 promoter) was maintained in DMEM supplemented with 10%FBS.
Biochemical / physiological behavior.
Mode of action: this product can inhibit translation by inducing misreading of m-RNA template in prokaryotes and inhibiting protein synthesis. Antibacterial spectrum: hygromycin B has inhibitory effect on bacteria, fungi and higher eukaryotic cells.
Warning.
The product of hygromycin B should be stored at a temperature of 2-8 °C, and its dry fixed form can be stored at a temperature of 2-8 °C for at least five years. It can be stable for 30 days at 37 °C. (for solution) if stored at 2-8 °C, the solution can remain stable for two years.
Points for attention in preparation.
The product was purified by ion exchange chromatography. Hygromycin B can be dissolved in water at a concentration of more than 50 mg/mL, or in methanol or ethanol. The solution should be sterilized by filtration rather than high pressure sterilization. In addition, it is reported that hygromycin B solution will lose its activity during freezing, and it can remain stable in cold storage, so freezing should be avoided.
