General description.
Folin phenol method is most commonly used to determine protein concentration by Lowry method. It is also used for the quantification of total phenols. In this method, the protein was pretreated with copper (II) in an improved biuret reagent (alkaline copper solution stabilized with sodium tartrate). The addition of phenolic reagents can produce chromophore which can increase the absorbance between 550750nm. In general, the absorbance at the peak (750 nm) or acromion (660 nm) is used to quantify the protein concentration between 1 and 100 mg/ml, while the absorbance at 550nm is used to quantify the higher protein concentration. In the absence of copper, the color intensity is mainly determined by the contents of tyrosine and tryptophan in the protein, and to a lesser extent by cysteine and histidine. Copper (II) has no effect on the color formed by tyrosine, tryptophan or histidine, but reduces color formation due to cysteine. Many changes to the original detection process have been published, including enhanced coloration, determination of insoluble protein content, and automation of the process. A variety of substances, including many buffers, chelating agents, detergents and cyclic organic compounds, can interfere with the detection of Lowry proteins.
