Application.
Used for cell separation and organelle separation.
Polysucrose 400 is used in conjunction with sodium pantothenate (Paque) to produce a poly-sucrose-sodium panchromate density mixture for the separation of cells and organelles, especially blood-derived lymphocytes and monocytes. It can be used in tissue engineering and macromolecular conformational research, and in the development, evaluation and use of macromolecular crowding (MMC) systems and structures. Polysucrose 400 can be used in the study of cell freeze drying.
Points for attention in preparation.
The concentration can reach 50% (w / v) in water. Under continuous stirring, polysucrose should be added slowly. The solubility of polysucrose 4001g in 10ml deionized water was tested by Abcone, and it was clarified to slightly turbid, colorless to light yellow solution. Polysucrose is stable in alkaline and neutral solutions. When the pH value is less than 3, it is hydrolyzed rapidly, especially at high temperature. Polysucrose can be sterilized under high pressure at neutral pH,110 ℃ for 30 minutes. The use of strong oxidants and reducing agents should be avoided.
Features and advantages.
Because of the high molecular weight and low content of dialyzable materials, the permeability of polysucrose to cell membrane is much lower than that of sucrose. Therefore, it can be expected that cells are collected at a lower density in the polysucrose gradient than in the sucrose gradient. Because of its low membrane permeability and low osmotic pressure, separation in polysucrose can usually better maintain cell function and morphology.

Poly sucrose 400 is a highly branched polymer formed by the copolymerization of sucrose and epichlorohydrin. Polysucrose 400 is completely Nonionic. Because it is rich in hydroxyl groups, polysucrose 400 is very hydrophilic and soluble in water. The most common application of polysucrose 400 is as a density gradient medium for the separation and isolation of eukaryotic cells, organelles and bacterial cells.
