Emur64 is soluble in water. 20mg / ml solutions can be prepared in water (which may require heating). The recommended reserve solution is 1mM aqueous solution. The effective concentrations used as protease inhibitors are 1 to 10 μ M. The aqueous solution is stable at-20 ℃ for several months. The diluted solution was stable in neutral pH for several days. Emur64 is stable in pH 2-10, but unstable in ammonia or HCl. Emur64 is also soluble in DMSO, and 10mM solution can be prepared in dry DMSO and stored at-20 ℃. Then diluted in the medium. The injection solution is prepared by dissolving Emur64 in 0.9% sodium chloride or a minimum amount of saturated sodium bicarbonate, and then diluting it with 0.9% sodium chloride (adjusting pH to 7.0 with acetic acid).
Application.
Emur64 is an effective ligand for affinity purification of cysteine protease. When coupled with mercaptan affinity matrix, the binding is no longer irreversible, but retains specificity.
Biochemical / physiological behavior.
Emur64 is an irreversible, effective and highly selective cysteine protease inhibitor. Emur64 does not react with non-protease functional mercaptan groups, such as L-lactate dehydrogenase or creatine kinase. EMel 64 does not inhibit serine protease (except trypsin) like other cysteine protease inhibitors, leupeptin and antipain. The trans epoxysuccinyl group (active part) of Emur64 irreversibly binds the active mercaptan groups of many cysteine proteases, such as papain, kiwifruit enzyme and cathepsin, to form thioether bonds. Emur64 is a very useful cysteine protease inhibitor for in vivo studies because of its specific inhibitory effect, permeability and low toxicity in cells and tissues.
