General description.
Cyclodextrins are cyclic oligosaccharides composed of 6, 7 or 8 glucopyranose units, usually called α -, β-or γ-cyclodextrins, respectively. These compounds have a rigid ring structure, making them natural complexing agents. The unique structure of these compounds is attributed to their stability of intramolecular hydrogen bonds between C2-and C3-hydroxyl groups of adjacent dextran units. The molecule is circular, C2-and C3-hydroxyl groups are located around the larger opening, and the more reactive C6-hydroxyl group is arranged around the smaller opening. The arrangement of C6-hydroxyl groups opposite to the hydrogen-bonded C2-and C3-hydroxyl groups forces the oxygen bonds to approach closely in the cavity, resulting in an electron-rich hydrophobic interior. The size of the hydrophobic cavity is a function of the number of dextran units forming cyclodextrins. The solubility of natural cyclodextrins is very poor. In the late 1960s, it was found that the chemical replacement of the 3 and 6 hydroxyl sites of 2BI greatly increased the solubility. Most chemically modified cyclodextrins can reach a concentration of 50% (w / v) in water. Pore size is the main determinant of which kind of cyclodextrin is used for complexation. The cavity diameters of β-cyclodextrin or β-glucopyranose unit compounds are ideal for use with many molecules commonly used in hormones, vitamins and many compounds in tissue and cell culture. Therefore, β-cyclodextrin is the most commonly used complexing agent.
Application.
Used to improve the solubility of non-polar substances such as fatty acids, lipids, vitamins and cholesterol in cell culture applications.
