Biochemical / physiological behavior.
Benzamidine hydrochloride is a reversible inhibitor of trypsin, trypsin-like enzyme and serine protease. About the concentration of 1mM can inhibit the common proteases. To inhibit protease from yeast, the concentration ranges from 0.5 to 4.0mM, and it is usually interchangeable with pepsin inhibitors.
In addition to being used as a strong competitive inhibitor of trypsin, benzamidine hydrochloride can also be used as a strong competitive inhibitor of thrombin and fibrinolytic enzyme. It was also found that benzamidine hydrochloride was as effective as aprotinin in preventing the degradation of glucagon in human plasma.
Peptidase inhibitors.
Points for attention in preparation.
Benzamidine hydrochloride is soluble in water and ethanol. When the concentration is measured in water, the clarified solution can be obtained by heating during 50mg/ml. Benzamidine hydrochloride is easy to be oxidized. It is recommended to prepare fresh solution in degassed water before each use. However, frozen equivalents stored in inert gases to remove air can remain stable for a short period of time. Insufficient information is available to evaluate the shelf life of frozen solutions.
Application.
Benzamidine hydrochloride hydrate has been used to:
Western blotting analysis.
Immunoprecipitation.
Purification of yeast dynamic protein.
As a protease inhibitor, it is used to prepare brush marginal membrane vesicles.
Deal with the sleep of synovial fluid, blood and eyes.
Prevent non-specific degradation of cartilage
